Autistic Children May Have Too Many Brain Cells

TUESDAY, Nov. 8 (HealthDay News) -- The brains of autistic children have far more neurons in the prefrontal cortex than the brains of kids without autism, finds a new study that could advance research into the disorder.

"For the first time, we have the potential to understand why autism gets started," said study author Eric Courchesne, a professor of neurosciences at the University of California, San Diego School of Medicine and director of the Autism Center of Excellence.

"Creating brains cells and the correct number of brain cells is absolutely fundamental to building the brain," said Courchesne. "If there is an excess number of neurons, there must be a negative consequence to that in the way the brain gets wired or organized."

In this small, preliminary study, the researchers examined postmortem brain tissue from seven boys with autism and six boys without autism who were aged 2 to 16 when they died.

The autistic children had on average 67 percent more neurons -- a type of brain cell and a fundamental building block of the nervous system -- than boys without autism of a similar age.

Specifically, they found autistic children had 79 percent more neurons in the dorsolateral prefrontal cortex and 29 percent more in the mesial prefrontal cortex than other kids.

The prefrontal cortex is key to complex thoughts and behaviors, including language, social behavior and decision-making. The dorsolateral prefrontal cortex is closely linked with "executive function," including planning, reasoning and "very high level cognition," said Lizabeth Romanski, an associate professor of neurobiology and anatomy at the University of Rochester Medical Center, who was not involved with the research. The mesial prefrontal cortex is thought to be important to social and other behavior and emotions.

While typically developing kids had about 1.16 billion neurons in the prefrontal cortex, autistic children had about 1.94 billion.

The study is published in the Nov. 9 issue of the Journal of the American Medical Association.

Autism is a neurodevelopment disorder characterized by problems with social interaction, communication and restricted interests and behaviors. An estimated one in 110 U.S. children -- many more boys than girls -- has the disorder, according to the U.S. Centers for Disease Control and Prevention.

Neurons are generated only during fetal development, specifically between 10 weeks and 20 weeks, Courchesne said. While neurons continue to grow in size during childhood, and brain connections get built and pruned, the number of neurons remains constant from birth.

That means that whatever goes wrong in autism starts in utero, which should help focus researchers looking for its causes or triggers, including specific genes or prenatal exposures.

"Now let's find out what genes or what in-utero, non-genetic conditions lead to an excess number of neurons," he said.

Prior research has also documented "brain overgrowth" in autistic children, but those studies were done by measuring brain circumference or MRIs, experts said. In this research, researchers were able to be more specific by counting brain cells in the prefrontal region.

"This very nicely builds on previous research and tries to explain why this increase in brain size might be, and what they find is it's because of an increased number of neurons," Romanski said.

After a period of proliferation during the second trimester, neurons are also "pruned," meaning that they undergo a planned cell death. "This pruning process, the dying off of cells, is a very important part of brain development," Romanski said.

One question that needs to be explored is whether autistic brains generate more neurons, or if they have a malfunctioning "pruning" process, she said.

Nicholas Lange, an associate professor of psychiatry and biostatistics at Harvard Medical School in Boston, cautioned that the study was small. He also said more needs to be learned, including whether excess neurons in the prefrontal cortex occur only in autism or in other developmental conditions, or even in any typically developing kids, as well.

Some of the kids with autism had many extra neurons, but not all had brains out of the normal range for weight, as would be expected. "The relationship between increased neuron count, brain overgrowth, and increased brain weight in autism is complex," Lange wrote in accompanying editorial.

Conducting postmortem brain tissue studies is a lengthy process because there are few brains available to study, Courchesne said. Eight of the 13 children whose brains were studied had drowned.

"So very seldom do people at that moment make the decision to donate their child's brain tissue for research, and in the absence of brain tissue for research, it goes very slowly," he said.

    This is a very interesting research. Hopefully we will able to learn about autism and other conditions, and debunk myths that society believes to be true, which leads to dangerous decisions. Unfortunately, I really don't believe society will ever listen to this. They are only focused on their personal goals instead of trying know more about the world. This pattern of thinking will eventually lead to a large disastrous result, which may occur this year.

    Does nobody ever pay much attention to the ideas that Socrates and Plato demonstrated? Though perhaps basic or ancient, their values still hold true today. Try to understand the reason and explanation for every action,event or decision that happens in the world, then make a better, informed decision. Even if it involves politicians, ask yourself that question, regardless of their position.

    This is fascinating & fantastic! I always felt my son's Autism was neurologic-genetic based-that he was "off" right from the start, NOT all of a sudden regressing after immunizations & he never had ear infections. I always describe his Autism "as if his brain is constantly on fast-forward & it's beyond his control, not behavior based". Let's keep the research going in this area!

    To those who point to evolution:

    Evolution is a slow process. Autism has increased by 600% in the last 20 years. In the 1960's the rates were 1 in 10,000 today 1 in 70 American boys has autism according to the CDC. This is more likely mass poisoning. Hard to admit this could happen in the US.

    I want our gov't to investigate the counterfeit pharmacueitcal drugs being imported from China to the US. For background on the problem, search online and read: "A Toxic Pipeline" New York Times series about the toxic counterfeit and often mislabled drugs being exported from China and watch "The Panama Syndrome" also from the New York Times.

    Chinese drug counterfeiters cover their trail by shipping through free trade zone like Dubai, UAE. In Panama, hundreds of senior citizens and young children were killed or seriously injured by counterfeit cough syrup from China.       Slow in general yes, but can also happen very rapidly.

      For example you can see a sudden increase in diversity and variance within a species (a lot of mutations in other words), but since most of them do not make it in the long run, a few years later it will look like there has only been one or two mutations (the rest died).

      What about the parents examining their drug use? When you indulge in bad habits like pot smoking starting during your teen years and drinking, and other drugs, you are altering the genes that you pass on to your kids. That's something that too many people just don't get - your body responds to everything you do, drink and eat - and if you use chemicals, your brain and your entire body including your reproductive system is going to be affected. Why do you think these skinny Hollywood women can't get pregnant? Years of starving themselves - Nature will not let them get pregnant becuase they have no fat stores to support the fetus.

      Evolution can and does happen within several generations. Since survival of the fittest isn't the norm for humans anymore (due to good nutrition, medical care), many genetic anomalies are perpetuating and you see it in future generations. Look at autism as an example. Someone commented here that they have several austistic people in their current family, and a few "quirky" people up the family tree. If this is true for other families, then the gene that is responsible for autism has been handed down through the branches of the famly tree, causing the huge increases you see today. This is true for ADHD, as well. That's my theory for what it's worth!

    I'd like to see this go one step further and see if there is a difference between children with birth onset autism and children with regressive autism. My son is definitely a genetic case - there's been two other cousins diagnosed with Aspergers in the last two years and we've traced several "quirky" family members up the family tree. But is it the same as the children who progress normally until 18-24 months then regress?

      Excellent point. My oldest showed Asperger's traits in his crib at 3 months. The second oldest developed normally and then went backwards into high-functioning autism. Both very smart and very loving, and very different boys.

      I think it is being reported and discussed amongst politicians, actually. There is a lot of research being done. There are many documentaries out there about autism. It probably has many different causes, and therefore they are very difficult to prove. Just like we don't know what causes many cases of cancer...

Because.many politicians get generous campaign contributions from Pharma.. they don't want to bite the hands that feeds them. The same holds true for the mainstream media who rely on lucrative drug ads..Have you watched the Nighty News lately? They are laced with drug commercials..Pharma has bought the politicians and and news media in this country.

      BTW, on World Autism Day last April, the White House declined to light the White House up blue (the chosen color for autism) in support of autism families...what a slap in the face!! A simple gesture would mean so much to our families. They lighted the WH up pink for breast cancer so why not blue for autism? Obama who I actually voted for on the first round will not get my vote..

      His administration has not been helpful. Kathleen Sebellius is useless. She ignores us. Parents are left to help eachother

    Disgusting to see this autism became an epidemic and basic studies like these are performed only in 2011. No matter the excuses....
    just a shame, to these many little kids with autism being non verbal and closed up in their own world. How many stories of parents child suicide have we heard of.

    Unfortunately, articles like this don't seem to resonate with a large portion of the electorate. The answers about autism will only be resolved through scientific research, yet many politicians are "anti-science" in their speech and deeds. Those who want to cut education are the among the biggest villains because they are snuffing out scientific research at its roots. We should be increasing funding for education and research, not cutting it.

    Yes! And the crowding of neurons results in faster reaction times, though tempered by over-thinking. Asked a seemingly simply question - What's your favorite actor - and while many would just blurt out some heart throb, their thoughts go from - Do I have a favorite? - Should I have a favorite - What makes someone a favorite - Should I choose based on a role, or on their skill? - Should I choose based on their looks? - etc, etc. All in just a second or two, but to the questioner, they seem to be uncommunicative - Autistic!

    Wonderful discovery - glad to hear they are untangling some of the spaghetti-tangle of symptoms. So tired of hearing people have to defend the reality of autism, even to their kid's doctors! There are still people who think autistics are either mentally challenged or just not 'disciplined' properly by their parents. Read a book, "One, Two, Three" (by Eleanor Craig) written a good while back, great insights for those who don't understand.

    This is incorrect. Life-long neurogenesis is now a fact and accepted by leading neuroscientists in mainstream neuroscience. This author either has misunderstood the scientist responsible for the study or the scientist is an old-timer who has not kept up with the times. Neurogenesis occurs throughout life in several small areas of the brain including the hippocampi.

    It's amazing how much misinforation exists on the web - even in news articles!!! If you would like a great perspective on the fundamental changes in neuroscience that have occured over time, read Norman Doidge's The Brain that Changes Itself. I promise you will not be disappointed. It's a brilliant book based on cutting-edge scientifc research and proves that our brains are far superior at changing than we ever thought (obviously it's more difficult later in life, but it is possible).
